چکیده

The effect of heavy metal ions (Cd, Cr 3+ , Cu 2 , Ni, Pb 2+ and Zn) on the growth and germination of conidia of the aphid pathogenic fungus Pandora neoaphidis was studied. The metal ions were added into the culture medium in three concentrations: A – concentration corresponding to the mean content of that metal in Polish soils, B – concentration 10-times higher and C – 100-times higher than the mean ones. The investigated heavy metal ions, except for Ni, added to the media at the concentrations corresponding to the mean content of these metals in Polish soils, did not affect the growth of aphid-pathogenic fungus Pandora neoaphidis. Ni, Cu, Zn and Cr, added to the media at a concentration 100-times higher than in Polish soils, prevented the growth of the pathogen. All the tested metal ions, except for Cu, added to the media at the mean soil concentration, did not affect conidial germination of the fungus. The conidia of P. neoaphidis were unable to germinate in the presence of the Cr, Cu, Pb and Zn ions in the medium at a concentration that was 100-times higher than the mean one. Cu and Zn caused a significant reduction of conidial germination even at a concentration that was 10-times higher than the mean content of these metals in Polish soils. This work suggest that strong pollution of soil by some heavy metals could be a restrictive factor of development and pathogenicty of entomophthoralean fungi in the environment.

Intraspecific variability of biological characteristics within entomopathogenic fungus Pandora neoaphidis was evaluated. Fifteen isolates of the fungus were obtained from 5 aphid species in Slovakia. Size of conidia, conidial germination, virulence, radial growth, and biomass production were evaluated. Conidial size varied considerably with exception of isolates originating from the same host p...

First report of Pandora neoaphidis resting spore formation in vivo in aphid hosts.

The entomopathogenic fungus Pandora neoaphidis is a recognized pathogen of aphids, causes natural epizootics in aphid populations, and interacts and competes with aphid predators and parasitoids. Survival of entomophthoralean fungi in periods of unsuitable weather conditions or lack of appropriate host insects is accomplished mainly by thick-walled resting spores (zygospores or azygospores). Ho...

Selection of reference genes for expression analysis in the entomophthoralean fungus Pandora neoaphidis

The selection of suitable reference genes is crucial for accurate quantification of gene expression and can add to our understanding of host-pathogen interactions. To identify suitable reference genes in Pandora neoaphidis, an obligate aphid pathogenic fungus, the expression of three traditional candidate genes including 18S rRNA(18S), 28S rRNA(28S) and elongation factor 1 alpha-like protein (E...

The Endosymbiont Arsenophonus Is Widespread in Soybean Aphid, Aphis glycines, but Does Not Provide Protection from Parasitoids or a Fungal Pathogen

Aphids commonly harbor bacterial facultative symbionts that have a variety of effects upon their aphid hosts, including defense against hymenopteran parasitoids and fungal pathogens. The soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is infected with the symbiont Arsenophonus sp., which has an unknown role in its aphid host. Our research goals were to document the infection fre...

Identification and validation of reference genes for qRT-PCR studies of the obligate aphid pathogenic fungus Pandora neoaphidis during different developmental stages

The selection of stable reference genes is a critical step for the accurate quantification of gene expression. To identify and validate the reference genes in Pandora neoaphidis-an obligate aphid pathogenic fungus-the expression of 13classical candidate reference genes were evaluated by quantitative real-time reverse transcriptase polymerase chain reaction(qPCR) at four developmental stages (co...
